The Experimental and Commercial Release of Transgenic Crop Plants   总被引:1,自引:0,他引:1  
With advances in recombinant DNA methods and transformation procedures, it is possible to transfer genes into crop plants from unrelated plants, microbes and animals. Many of the modifications being carried out, or envisaged, are for disease and pest resistance, product quality and tolerance to environmental stress, but there are additional opportunities to modify crops to give specialized products for industrial or pharmaceutical use. Some of the characteristics of transgenic plants are considered, including: transgene copy number, position, expression, stability, pleiotropy, selectable marker genes and somaclonal variation. There have been several hundreds of field trials with transgenic plants, and the first transgenic varieties are likely to be approved for commercial production in 1993. Before releasing transgenic plants, it is necessary to carry out a risk assessment to determine whether the transgenic variety will behave differently from a conventionally bred variety. Assessment procedures are being harmonized internationally by various organizations. There is a growing commitment to apply these genetic modification methods to crops in developing countries, as genes relevant to their crops and environments become available.  相似文献

Summary Many aspects of basic and applied problems in plant biology can be investigated by transformation techniques. In dicotyledonous species, the ability to generate transgenic plants provides the tools for an understanding of plant gene function and regulation as well as for the directed transfer of genes of agronomic interest.For many dicotyledonous plants Agrobacterium tumefaciens can be routinely used to introduce foreign DNA into their genome. However, cereals seem to be recalcitrant to Agrobacterium-mediated transformation.In cereals, many efforts have been made in recent years to establish reliable transformation techniques. Several transformation techniques have been developed but to date only three methods have been found to be suitable for obtaining transgenic cereals: transformation of totipotent protoplasts, particle bombardment of regenerable tissues and, more recently, tissue electroporation. The current state of transformation methods used for cereals will be reviewed.  相似文献

Transformation efficiency of melon is low and is still regarded as a challenge. In this paper, the regeneration and transformation response of ‘BU‐21/3′, a newly characterized melon breeding line, is described. The line seems to be superior in this regard to previously evaluated genotypes. Agrobacterium‐mediated delivery of the GUS or GFP reporter genes into cotyledon explants was used to evaluate efficiency of transient and stable transformation. Good transient expression was observed, and stable transformation frequencies of 0.4‐1.5 transgenic shoots per explant were obtained. Transgenic plantlets were transferred to a contained greenhouse as early as 8‐10 weeks after transformation. Transgenic plants are fertile and exhibit a true‐to‐type phenotype. The ‘BU‐21/3’ line may become a useful tool for the facilitation of transgenic breeding in melon.  相似文献

Summary Pollen grains of 14 Cucumis accessions were irradiated with 0, 1, 2, 3 or 4 kGy acute gamma rays and germinated in vitro directly afterwards. Pollen germination was significantly reduced by increasing irradiation dose for all species, except C. melo var. agrestis. Pollen tube growth was generally reduced likewise. Pollen of two C. anguria subspecies was most sensitive to irradiation. Sensitivity of the pollen with respect to pollen tube growth in relation to irradiation dose was inversely related to total DNA amount per nucleus. In vitro germination was not related to DNA amount per nucleus. Results show that the examined Cucumis species, especially C. melo var. agrestis, are sufficiently resistant to irradiation to be used as donor species for in vivo egg cell transformation of the cucumber.  相似文献

Summary The merits and limitations of somatic cell techniques involving Agrobacterium-mediated transformation, direct gene transfer and protoplast fusion, are discussed in relation to the genetic improvement of forage and grain legumes. Whilst progress with legumes is limited compared to that with plants of other families such as the Solanaceae, the fact that many legumes are readily amenable to tissue culture now permits somatic cell techniques to be targetted to these species. Future development of the subject will necessitate close collaboration between molecular biologists and plant breeders to enable novel plants generated by in vitro technologies to be incorporated into conventional breeding programmes.  相似文献

侵蚀破坏土壤团聚体结构,使团聚体破碎产生更小的可迁移颗粒而导致土壤养分的运移;侵蚀还改变了土壤的水分状况,水蚀在一定程度上增加了土壤的含水量,风蚀则可使土壤的含水量降低。团聚体结构的破坏以及土壤水分状况的改变与土壤的氮素迁移转化过程有着密切的联系。本文仅对土壤侵蚀造成的土壤物理性状的改变与土壤氮素迁移转化作一概述,并就耕作、覆盖等降低土壤侵蚀的农业措施与土壤氮素转化的关系进行阐述,提出了侵蚀土壤氮素研究的去向。  相似文献

通过根癌农杆菌介导法将甜菜碱醛脱氢酶(BADH)基因导入马铃薯栽培品种甘农薯2号, 经PCR、Southern杂交和Northern杂交证明BADH基因已整合到马铃薯基因组中并在转基因植株中转录和表达。测定表明对照植株没有BADH酶活性, 各转化株系在胁迫前后BADH酶活性近似, 在2~11 U之间。BADH酶活性与叶片的相对电导率呈一定的负相关(y= –3.7738x+57.083, r=0.989**)。在NaCl和PEG胁迫下, 转基因植株生长正常, 株高比对照提高0.41~1.00 cm, 单株重量比对照增加10%~35%, 说明外源BADH基因的导入提高了马铃薯植株对干旱和盐碱的抗性。  相似文献

It is difficult to establish precise mathematical model in extended Kalman filter data fusion algorithm for GPS/ DR integrated vehicle navigation system, an improved extended Kalman filter data fusion algorithm is put forward based on Discrete stationary wavelet transformation.
